Effects of Salinity increase on some Blood factors in juveniles Rainbow trout (Oncorhynchus mykiss)

This study aims to find the changesin blood factors in 150 Rainbow trout fingerlings with 20.01±1.33 average weight. Fish were studied in different salinity levels including 7ppt, 11ppt, and 0ppt (fresh water) with 3 replicates for each ones. Blood sampling was performed from caudal vein at times 0, 24, 48, 72, and 240 hours after the beginning of experiment. After centrifugation and separation of blood plasma, Hemoglobin concentration, hematocrit percent, number of white and red blood cells, lymphocytes, monocytes and neutrophils percentages was measured in blood. Results showed that exposure to high salinity, reduced the hematocrit percentage and in times 24 and 240 compared to zero time (fresh water), significant differences were found (P< 0.05). But during the experiment the hemoglobin concentration did not show significant differences with time zero(P>0.05).The number of white blood cells at 24 h and 11ppt salinity had a significant reductioncompared to zero time(P<0.05). The number of red blood cells in 11ppt salinity and 240h reached its lowest level during the test(P<0.05).In examining the percentage of lymphocytes and monocytes, it was found that at all times and in all salinity levels, the highest percentage was owned by lymphocytes meanwhile regarding their percentage no significant differences was found between experimental and control groups(P>0.05). Percentage of neutrophils, in 7ppt salinityand 72h, had significant differences with the other times and at the end of 10th day,didn’t show any significant difference with them statistically (P>0.05).
