Molecular Identification of Some species of Serranidae Family in Qeshm and Hormuz Islands using COI gene sequence

Molecular identification of economic fish species of the Persian Gulf, including grouper fishes, seems necessary in order to protect the resources of native species. For this purpose, in this study, three species of grouper fish species including Epinephelus coioides, Epinephelus radiates and Epinephelus areolatus were sampled from the Qeshm and Hormoz islands in the summer of 2022. DNA extraction was done and a part of the mtDNA genome (cytochrome oxidase I or COI) was amplified using PCR method. The phylogeny analysis was done using Neighbor Joining model based on the genetic distance of the studied species. The phylogeny results showed that the samples belonging to E.radiates and E.coioides species were placed in the first clade in two different clusters. In this regard, the E.coioides sample was placed in the first cluster together with samples from Qatar and Pakistan and also samples of the same species from the Persian Gulf. In the second cluster, E. radiatus species were placed together with samples from India, Taiwan, China and Canada. Specimens of species E. areolatus with samples from India and Egypt were placed in the second clade. The phylogenetic analysis indicated the identification of a new haplotype of E. radiatus species with significant genetic differences from other samples registered in the world gene bank (NCBI). This was while the sequence of E. radiatus species had not been reported from Iran. In this regard, E. radiatus species can be considered as a new and native species of the eastern region of the Persian Gulf. The results of mitochondrial DNA analysis, confirmed the significant genetic divergence between the studied species. The results of this study has an effective role in sustainable exploitation, protection of biological diversity and fisheries management of grouper fishes.
